i would like to prepare a spike sample how do i prepare it ? for example the preparation of sample in the method is to add 2g of sample into 20ml volumetric flask , so if i want to prepare a spike how do i prepare it , the amount of the weight ? ( i was told to add 1g of known material into 4g of sample to reach 5g ( spike + sample ) to 50ml volumetric flask ( solvent is water ) and injected to HPLC with dilution factor of 10 ) i dont understand how they choose the specific weight and the calculations please explain in details from the preparation till the calculations .
