The enzyme alcohol dehydrogenase (ADH) carries out the conversion of alcohol to acetaldehyde, introduced at the beginning of chapter. NAD+ NADH NAD+ NADH ADH ALDH CH3CH2OH CH3CHO CH3COOH Ethanol Acetaldehyde Acetic acid. If you accidentally ingest methanol (CH3OH) instead of ethanol, the enzyme ADH produces toxic FORMALDEHYDE, which is in turn converted by the next enzyme in the pathway, aldehyde dehydrogenase (ALDH), into FORMIC ACID. Formic acid is especially toxic and strongly affects the optic nerve, so methanol poisoning can cause blindness. The treatment for methanol is, perhaps surprisingly, to give the patient ethanol (regular grain alcohol). The ethanol inhibits the ADH reaction that is converting the methanol to formaldehyde and formic acid. Here are the structures of methanol and ethanol: CH3OH CH3CH2OH Methanol Ethanol A) In the reaction above from ethanol to acetaldehyde, NAD+ NADH serves as a(n): (choose one) i) enzyme coenzyme iii) inorganic cofactor B) In this case, does ethanol act as a competitive or noncompetitive inhibitor of ADH? Justify your answer. C) Explain why ethanol treatment works to stop the toxic effects of methanol poisoning.

Metabolism of alcohols Now consider the metabolism of ethanol by ADH. Review the Aldehyde Oxidation Tutorial before you proceed. Then use the information you've learned so far to determine how ADH and ALDH act to metabolize different alcohols. Ethanol Feel free to refer back to the Alcohol Oxidation Tutorial as you progress. ADH metabolizes ethanol to which compound? No reaction occurs and no product is formed.
