A. Buffer Preparation - Prepare 10 mL of the following buffers by diluting the stock solutions in H2O (listed below):
1) Hypotonic lysis buffer:
- 5 mM HEPES [pH 7.9]
- 1.5 mM MgCl2
- 10 mM KCl
- 1 mM beta-mercaptoethanol (BME)
2) Nuclear Extraction Buffer:
- 20 mM HEPES [pH 7.9]
- 25% glycerol
- 1 M NaCl
- 1.5 mM MgCl2
- 0.5 mM EDTA
- 1 mM BME
3) SDS Lysis Buffer:
- 500 mM NaCl
- 1% SDS
- 1 mM BME
Stock Solutions:
- 5 M NaCl
- 1 M HEPES [pH 7.9]
- 750 mM MgCl2
- 1 M KCl
- 1 M BME
- 50% glycerol
- 100 mM EDTA
- 10% SDS
