TMP/SMX has two components that act on different steps in the folic acid biosynthesis pathway and are thus used together. Sulfamethoxazole is a structural analog of a compound called PABA, competing with it in the enzymatic production of dihydrofolic acid, an intermediate in the synthesis of folic acid.
