Statistical Review: Behavioral studies at adulthood Marble burying and nestlet shredding test The marble burying test showed that adolescent exposure to JWH-018 increased the number of completely covered mar- bles compared to vehicle group (((8)=2.51; p=0.036; +33%, Cohen' d=1.5874; Fig. 3(a)). Consistently, nestlet shredding test showed that adolescent exposure to JWH-018 increases nestlet shredding compared to their vehicle counterparts (((8)=2.97; p=0.018; +140%, Cohen' d= 1.8767; Fig. 3(b)). These data suggest that JWH-018 self-administration dur- ing adolescence exerted permanent, long-lasting repetitive/ compulsive-like behavioral effects at adulthood. The study used the SCRA reference compound, JWH-018, through means of intravenous self-administration (IVSA) in adolescent male mice and then evaluated behavioural effects in adulthood of these mice. All of the subjects tested for this study were CD1 adolescent male mice, weighing 22-27g. The mice were housed in standard conditions, in groups of three to six per cage prior to the IVSA experiments. At adulthood, after a drug-free washout period of approximately three weeks, the mice were randomly divided into different groups to undergo behavioural tests. The results found long-lasting repetitive/compulsory-like behaviour effects, such as marble burying and nestlet shredding in adulthood of mice which used JWH-018 through IVSA during adolescence [ CITATION Giu22 \l 3081 ]. The response variable is the behavioural effects at adulthood; marble burying and nestlet shredding in mice that have used JWH-018 in adolescence. Positive association is depicted as when the explanatory variable (JWH-018 use) increases, the response variable increases. The null hypothesis (H0) is that the behavioural effects at adulthood, such as nestlet shredding, is the same whether the subjects were administered the JWH-018 as an adolescent or not. The p-value for nestlet shredding is p = 0.018. This p-value suggests that there is moderate-to-strong evidence that the subjects that were subjected to the JWH- 018 during adolescence were more prone to behavioural effects in adulthood, such as nestlet shredding, than those who were vehicle.
