The bacteriophage commonly known as the phage, are viruses that infect bacteria. A virus is nucleic acid molecules that are surrounded by a protein coat. A virus reproduces inside of living cells, in this case the bacteria cell. CRISPER is an acronym for clustered regularly interspaced short palindromic repeats. CRISPR is a part of prokaryotic organism's resistance/defence against viruses. It has recently been involved in research for human genome editing. CRISPR creates immunity through three major steps. First acquisition where integration takes place, next expression of the proteins, and lastly interference. The cell lab provides a relation between bacterial resistance and phage immunity. This lab provides the student with either an example of a bacterial lawn or a clear plate found in the tested cultures. A bacterial lawn is an example of when the bacteria is immune to the phage, and a clear lawn demonstrates non-immune bacteria. In the lab there are two putative BIMs being tested for their phage immunity. Which putative BIM will have the longer DNA length, and why? This question will be actively answered throughout the lab as the BIM's are run through PCR using two different primers called CR1 and CR3 as well as the CRISPR regions. Through these experiments, the principle finding would be that BIM 1 in addition to 2 will travel similar lengths, however we see a greater change through the CR1 primer. 1b.
