How conditional mutagenesis screens can be used to find the role and function of genes in the cell cycle Nobel prize awarded in 2001 (below) for work done on the cell cycle of yeast. Leland Hartwell identified temperature sensitive mutants that don't progress through the cell cycle at the restrictive temperature - Saccharomyces cerevisiae - budding yeast - brewer's yeast . The large cells in the image below are parental cells. They bud off cells by cell division, which get bigger & bigger as those cells divide. Nurse Prize share: 1/3 The Nobel Prize in Physiology or Mediche 2001 was awarded jointly to Leland H. Hartwell, Tim Hunt and Sir Paul M. Nurse "for their discoveries of key reguistors of the cell cycle" . The cell cycle has 4 important phases. The 2 most significant are M phase (mitosis) and S phase (when DNA replication takes place). There are also 2 G (gap) phases - G1 and G2. These are where the cell is pausing between DNA synthesis and separation (mitosis & cytokinesis) · The bud gets bigger throughout the cell cycle process. Hartwell looked for cell cycle mutants. He chose Saccharomyces cerevisiae bc at each stage of the cell cycle there are clear morphological differences between the cells at different stages. . He looked for mutants that arrested at each stage of the cell cycle (seen in black & white image below). Mutagenesis was a key tool in identifying cells that couldn't complete the cycle. . However, a problem w identifying genes that are required for the cell cycle is that those cells no longer go through the cell cycle if they carry that mutation. How would you identify genes that regulate the cell cycle? Nuclear migration DNA replication Bud emergence Spindle pole body duplication Spindle formation Some cdc Mutants Wild type cdc 8 G M Chromosome segregation nuclear division cdc 24 cdc 10 G1 Cytokinesis START Growth Cell cycle of Saccharomyces cerevisiae Mutagenesis identifies key genes that are required for the cell cycle LO1
· Hartwell identified cell cycle mutations by using a temperature sensitive screen. At the permissive temperature, mutants have the same phenotype as wildtypes (below they are seen progressing through the cycle in the same way). Doesn't matter if permissive temp is warmer or colder than you would normally grow wildtype cells - the important feature is that they have the same phenotype. How do you screen for temperature sensitive mutants? - G1 S G2 M wild-type - mutant At the permissive temperature the growth of all mutants is comparable to wild-type Temperature sensitive mutants have a wild-type phenotype at permissive conditions · When you transfer cells
