How did experiments with Neurospora crassa mutants support the one-gene, one-enzyme hypothesis?
Beadle and Tatum proposed that each gene coded for one enzyme in the metabolic pathway. They worked with a bread mold (Neurospora) that only needs sugar, salts, and the vitamin biotin. They isolated several mutants; one mutant could not synthesize the amino acid arginine because it lacked an enzyme required for a step in the pathway. The wild-type (WT) mold had genes for enzymes 1, 2, and 3, which allowed it to grow on minimal growth media, while the mutant molds could only grow when given a medium supplemented with pathway intermediates (ornithine or citrulline). The hypothesis proposed by Beadle and Tatum was that each gene coded for one enzyme in the metabolic pathway.
