The chromatin immunoprecipitation (ChIP) technique allows a researcher to determine if a protein of interest binds to a specific region of DNA. To perform the technique, cells are treated with formaldehyde to cross-link proteins to DNA. The cross-linked DNA-protein mixture is extracted and the DNA is sheared into fragments. The protein of interest is then pulled down along with any cross-linked DNA. After releasing the DNA, the researcher tests if the specific region of DNA is present in the pulled-down complex, which would indicate that the protein was interacting with this DNA sequence.
