A researcher is interested in the impact of Mycobacterium tuberculosis infection on mammalian host cells, and is also interested in measuring gene expression in M. tuberculosis following infection. If the investigator wishes to gain all of this information from M. tuberculosis infected mouse cells using one RNAseq study, what type of RNAseq would you advise they carry out, and why?

Step 1
- By using dual RNAseq, the researcher can measure gene expression changes in both the mammalian host cells and the M. tuberculosis bacteria following infection. - This approach provides a comprehensive view of the interactions between the host and pathogen, Show more…

Mycobacterium tuberculosis and Neisseria meningitidis can both cause life-threatening diseases. Imagine you are working in a microbiological laboratory and you receive a sample that could contain one of these pathogens. Explain what types of tissue could have been given to you for detection of either of these bacteria. Describe two laboratory assays that would be useful to determine which bacterium you are dealing with. Indicate which outcome of each assay would point to which bacterium. Explain what other test you would conduct before recommending the best treatment for the infection and why that is important.

Mycobacterium tuberculosis is an intracellular bacterium that causes tuberculosis. The $M .$ tuberculosis genome includes 17 genes encoding adenylate cyclase, whereas free-living bacteria typically have just one AC gene. Why might it be an advantage for $M .$ tuberculosis to generate high adenylate cyclase activity?
Tuberculosis (TB) is caused by a bacterium spread from person to person through the air. It affects the lungs, but can also affect the kidneys, brain, and spine. A simple diagnostic test for TB involves injecting antigen (from the bacterium that causes TB) under the skin and then waiting a few days for a reaction to appear. If a reaction occurs, a person has the TB infection (although the majority of people won't develop an active disease with symptoms). Would this test be useful in diagnosing TB in patients with AlDS? Predict whether patients with both AlDS and TB infection would have a positive skin test or not.
