An immunology researcher has developed a transgenic mouse line that has been engineered to have inducible inactivation of PLC-gamma. In these mice, PLC-gamma is inactivated in hematopoietic stem cells following RAG1 expression when the mouse reaches adulthood. Based on what we learned about in lecture, what phenotype or phenotypes would you expect to observe in these mice? Check all that apply. - Reduced number of naive T cells - Reduced number of naive B cells - Reduced number of plasma cells - Reduced number of CD4+ memory T cells

If a transgenic mouse express transgenic T cell receptors that recognize an antigen derived from Escherichia coli presented by a major histocompatibility complex class II (MHC-II) allele expressed by the transgenic mouse, what happens? a. The transgenic mouse will be perfectly healthy. b. The transgenic mouse is more affected by viral infections than wild-type mouse. c. The transgenic mouse is more affected by bacterial infections than wild-type mouse. d. The mouse has higher titers of antibodies against antigens derived from Escherichia coli than wild-type mouse. e. The mouse will increase its expression of the MHC class II allele on antigen presenting cells (APC).

Irradiation of mice with a dose of 600 rad of total body irradiation eliminates 95% of total lymphocytes from the spleen and lymph nodes and also eliminates all antigen-specific memory B cells. Nonetheless, when Influenza A-infected mice are subjected to this irradiation at 60 days post-infection, and then reconstituted with bone marrow cells from a naive mouse (this replenishes all of the lymphocyte populations), the levels of circulating anti-Influenza A IgG antibodies show nearly no decline when mice are monitored for the following year. What is the explanation for this finding?

Altered Metabolism in Genetically Manipulated Mice Researchers can manipulate the genes of a mouse so that a single gene in a single tissue either produces an inactive protein (a "knockout" mouse) or produces a protein that is always (constitutively) active. What effects on metabolism would you predict for mice with the following genetic changes: (a) knockout of glycogen debranching enzyme in the liver; (b) knockout of hexokinase IV in liver; (c) knockout of FBPase-2 in liver; (d) constitutively active FBPase- 2 in liver; (e) constitutively active AMPK in muscle; (f) constitutively active ChREBP in liver?
