Assignment 10: Antibody questions
Here is some information. Antibodies are themselves proteins, and the purified protein that is an antibody is called immunoglobulin-gamma (IgG). A local biotech company called Antibodies-R-Us makes antibodies for sale to the scientific community, and does so by extracting and purifying IgG from rabbits and then injecting the rabbit IgG into goats. After several weeks, they harvest IgG from goat serum (goat antibodies that recognize and bind to rabbit antibodies), they purify the goat IgG, and chemically bind the goat antibodies to one of several types of detecting agents (e.g. HRP enzyme, Cy2 fluorophore, or Cy5 fluorophore) for which they sell to the research community.
The protein that you want to detect in your research lab is called "Hammerhead" and you want to use an antibody to determine the location of where the Hammerhead protein resides in the cells of zebrafish neurons by visualizing Cy2 (Cy2 is a fluorescent labelling molecule). You don't have an antibody for Hammerhead yet, so you inject purified Hammerhead protein into a rabbit, wait a few weeks, give the rabbit a booster shot of Hammerhead, wait a few more weeks, then extract and purify from rabbit serum all IgGs that specifically target Hammerhead.
