Recently a vaccine that uses synthetic mRNA has shown promising results for generating a robust and protective immune response to the novel coronavirus of 2019 (SARS-CoV-2). This is a new vaccine technology that uses mRNA to produce antigenic viral proteins within our cells, as shown in the figure below. In the past, this technology was restricted by the instability and rapid degradation of the foreign mRNA. Several new improvements to the method have overcome this problem however, making it a viable and transformative technology in vaccine development. One of these is the pre-incubation of the mRNA with eukaryotic initiation factor 4 (elF4). A. Explain a plausible reason why elF4 could stabilise the mRNA in the recipient cell? B. The sequence was also scanned for known miRNA complementarity and was changed by alternative codon usage where this was found. Why is this necessary? C. What are some other modifications to the viral RNA that could be added to tie viral mRNA to aid stability in the recipient cell?
