The schematic depicts the complement activation pathways and the sites of inhibition by the various pharmacological compounds. j. Given what you know about the effector mechanisms of antibodies, comment on how they would function as therapeutics. (4) k. How would treatment with AMY-101, a C3 inhibitor, reduce inflammation in a COVID patient? (4) l. How would IFX-1 and Advoralimab help prevent CRS? (C5aR is the receptor for C5) (4)

Step 1
Antibodies function as therapeutics by binding to specific antigens, neutralizing them, and promoting their clearance from the body. They can also activate the immune system to destroy the target cells or pathogens. Show more…

Consider the structure of the immunosuppressant $F K-506,$ a molecule shown to disrupt calcineurinmediated signal transduction in $T$ -lymphocytes. (a) What is the molecular formula of this immunosuppressant? (b) How many stereocenters are present in $F K-506 ?$ Determine the maximum number of stereoisomers possible. (c) Identify and label the various functional groups present. (d) Consider the two stereocenters in this structure labeled with asterisks ("). Determine the absolute configuration of each stereocenter. (e) $F K-506$ has been shown to exhibit moderate solubility in various organic solvents. Is this immunosuppressant expected to be soluble in ethanol $\left(\mathrm{CH}_{3} \mathrm{CH}_{2} \mathrm{OH}\right) ?$ (f) Consider the carbon atom labeled "1." Describe the geometry and approximate bond angles about this carbon atom. (g) Draw the alternative chair conformations of the cyclohexane ring at the lower right of $F K-506$ and label the more stable conformation. (h) Are there any aromatic components present in $F K-506 ?$ (i) Patients taking $F K-506$ have reported several side effects from this medication, including headaches, nausea or diarrhea, and slight shaking. Would you expect the enantiomer of this drug to result in the same side effects?
You have prepared knockout mice with mutations in the genes that encode various complement components. Each knockout strain cannot express one of the complement components listed across the top of the table below. Predict the effect of each mutation on the steps in complement activation and on the complement effector functions indicated in the table using the following symbols: NE = no effect, D = process/function decreased but not abolished, A = process/function abolished. Complement component knocked out: C1q C4 C3 C5 C9 Factor B MASP-2 Formation of classical pathway C3 convertase Formation of alternative pathway C3 convertase Formation of classical pathway C5 convertase Formation of lectin pathway C3 convertase C3b-mediated opsonization Neutrophil chemotaxis and inflammation Cell lysis

Fluoroacetate is a poison used to kill pesky animals (rats, coyotes, etc.). Animals that ingest fluoroacetate have 10 times the normal cellular concentration of citrate and very low concentrations of isocitrate. What kind of inhibitor is fluoroacetate? Describe and draw out the pathway of fluoroacetate poisoning, showing the structures and names of all reactants and products, as well as the names of the enzymes. You do not need to draw the structures of ATP, NAD, CoA, etc., but do show them as reactants or products in the appropriate places. You do not need to show the mechanisms of the enzymes.
