It depicts the energy landscape of a protein as it folds from a denatured (unfolded) state to its native (folded) state. The funnel's wide top represents the many possible unfolded conformations, while the narrow bottom represents the single, low-energy native Show more…

The elegance and importance of proper protein folding and assembly cannot be overstated. Simple electrostatic interactions are fully responsible for long peptide chains becoming giant, globular, functional proteins. Research and discuss two examples of improper protein folding. What is the cause of each malformed protein? What is the consequence of this deformity? What are potential treatment options, and how do they address and remedy the problem? Include examples other than the two mentioned in the textbook.

Problem 4: Proteins can go from a disordered state, typically referred to as a random coil, to a highly ordered (folded) state. For many proteins, this process occurs spontaneously under biological conditions (i.e. inside cells). In fact, one common folding model is the "folding funnel" which describes how configurational entropy decreases from a large number of microstates in the disordered state to a small number in the folded state. Describe how these processes can occur spontaneously without requiring an external energy source even though the entropy of the protein decreases? (Note: the answer is not straightforward based solely on the material we learned, you may need to research the literature to find an accurate answer.). Statistical (Funnel) Model Disordered Intermediates disordered folded Folded Degrees of freedom
