During oxidative phosphorylation of cell respiration, H+ ions move through the membrane pumping electrons from one side of the membrane to the other. ATP is synthesized when H+ ions move through a protein channel provided by ATP synthase. ATP pushes electrons through membrane carriers creating a H+ ion gradient. a concentration gradient is generated when large numbers of H+ ions passively diffuse from the matrix of the mitochondrion to the mitochondrion's intermembrane space. a phosphate is transferred from an organic molecule to ADP.

Glycolysis produces ATP by phosphorylation of ATP unlike phosphorylation that is driven by the proton motive force. At the end of glycolysis, other than pyruvate and ATP, the molecule contains most of the free energy that will be used later to make more ATP. The electrons carried by FADH2 pump fewer into the intermembrane space because they transfer their electrons to the membrane protein of the transport chain.
