During initiation of DNA replication in a single bacterial plasmid, origin recognition protein binds at a single origin of replication. DNA helicase begins unwinding dsDNA in a GC-rich region. ssDNA binding proteins bind and activate DNA polymerase. All of the above (b&c).

Which of the following is/are true for prokaryotic and eukaryotic initiation of replication? (Select all that apply): The Origin recognition complex (ORC) is a six-subunit complex that acts as the first initiator of prokaryotic DNA replication and binds DnaA boxes. The initiator is a sequence of DNA that basically points at the actual start point for replication. The replicator is a protein that specifically recognizes a sequence in the replicator and activates the initiation of replication. The GATC methylation site in the origin of replication in E. coli is important as GATC methylations have fewer hydrogen bonds than non-methylated GATC sites. AT-rich regions of DNA in the origin of replication in E. coli are involved in regulating replication and are needed to ensure only one round of replication occurs. The conditions required for helicase loading onto DNA occur only in G1 of interphase, and the conditions required for helicase activation occur only in the S phase of interphase.
