For this lab on digestive enzyme activity, we will analyze the results for the carbohydrate digestion only (and not include fat and protein digestion reactions). The experiment is done in the following way: collect your saliva and mix with starch and inclubate mixture for one hour. There are four separate samples (see lab handout): 1) control without saliva, 2) with saliva, 3) in an acid environment (HCI), and 4) with boiling the saliva before adding the starch. Saliva contains the enzyme amylase that will break down starch into maltose. After the one hour of incubation, the samples are divided into two sets for testing. The first set is tested for the presence of starch (no breakdown) using Lugol's solution, the second set is tested for the presence of maltose, the product of amylase activity. Analysis the results in the two figures using the guidelines from the handout, what can you say about the optimal working conditions of the enzyme amylase? Is it needed for breakdown, is it inhibited by acid, or inactivated by high heat only? Answer each questions and explain them using the results shown from both the Lugol's test and the maltose test.
