You are interested in purifying the protein cytochrome from frogs, and you decide to use an antibody to assist you through immunoprecipitation. Unfortunately, you don't have an antibody for frog cytochrome. However, a collaborator of yours has an antibody for mouse cytochrome, which you think might work. How is it that an antibody for mouse cytochrome might work for this application? (In other words, what is likely common between mouse cytochrome and frog cytochrome that will allow an antibody to cross-react as you hope?) When you ask your collaborator if you can have some of her mouse cytochrome antibody, she asks you whether you want her monoclonal or her polyclonal version of the antibody. Which do you think would be more likely to work for your application, and why? You use the antibody in an attempt to purify frog cytochrome from frog cell lysate using immunoprecipitation. You isolate protein off of the antibody and run the protein elution in 2-D electrophoresis and stain the gel with Coomassie Blue. The data are below. Was your protein sample pure for cytochrome c? How do you know? How many different proteins were in your protein sample? kDa ladder Look up some information about cytochrome (not cytochrome oxidase).
