Disc Diffusion Assay is a method used to test for antimicrobial susceptibility. A disc treated with highly effective disinfectant, such as bleach, is usually used as a positive control, while untreated disc is used as a negative control. Why do we need a positive control in this method? Bleach is known to have no effect on the bacteria than the drug, so it shows what too large what its response looks like. Placing three different discs in one plate lets you take an average zone of inhibition measurement. The untreated disc shows that regular, uninhibited bacterial growth looks like around the disc. None of the options is correct. Positive control shows how an inhibition looks like.
