Experimental Background for Lab: Imagine that you have been hired by a small startup company called "Enzyma". Enzyma specializes in the discovery of enzymes with potential uses in the food and brewing industry. A researcher at Enzyma isolated an amylase from the ubiquitous soil fungus Aspergillus oryzae (ass-per-jill-cus or-eyec-zee) from the tropical rainforest in South America. The researcher has a good reason to believe that this enzyme might have superior capabilities to be used in the brewing industry where high maltose yields are desirable. You (and your group members) are part of the R&D (research and development) team at Enzyma, and your task is to figure out the optimal conditions for maltose production resulting from starch hydrolysis using the amylase isolated from A. oryzae. To quantify the amount of maltose present in an experimental situation, you will complete three tasks: 1) Create a standard curve of known maltose concentrations and measure the absorbance of those concentrations. 2) Determine the optimal pH conditions for the enzymatic hydrolysis of starch. 3) Finally, your boss would like you to determine the best temperature at which the enzyme will function. You will write hypotheses and design an experiment to answer your boss's question and get the promotion. Remember that a standard curve is like a ruler, a best-fit line, that you will use to measure something at a later time. Thus, you want to have known concentrations of the reagent to be quantified in your experiment. Here, maltose, either added or created by amylase, is the solute of interest. You will create the solutions necessary to create the standard curve using known concentrations of maltose that will reduce the DNS reagent, resulting in a change in coloration. The rest of the reaction volume will be comprised of water (solvent) and starch (substrate for the enzyme).
