8. Fig. 22.1 shows a transverse section of the stem of a typical pondweed viewed using a × 10 objective lens. Part of a graticule is shown below the stem. The markings on the graticule are 0.1 mm apart. vascular bundle air space A B 0 10 20 30 Fig. 22.1 (i) Measure the width of the stem between points A and B. Give your answer to the nearest 0.1 mm. You are using a new microscope for the first time, and need to calibrate the ruler (micrometer) that is inside the ocular lens in order to accurately measure any specimens. When using the 10x objective lens with the 10x eyepieces, you determine that a distance of 500 micrometers on a stage micrometer is equivalent to the distance measured by 40 units in your ocular micrometer. a. What would be the distance measured by 1 unit of your ocular micrometer, using these lenses? b. You then swap the stage micrometer slide with a slide of beautiful diatoms (photosynthetic single celled organisms with intricate silica cell walls). You measure one diatom to be 8 ocular units long using the same arrangement of lenses as above. How long is this diatom in micrometers?

Match the type of microscopy with the best corresponding description provided below. Each answer may be used once or not at all. Uses an optical component to take advantage of the different refractive indices of light passing through different regions of the cell. Produces a halo effect around the object. Has the best resolution and greatest magnification but produces a two-dimensional image. Requires the use of two sets of filters. The first filter narrows the wavelength range that reaches the specimen and the second blocks out all wavelengths that pass back up to the eyepiece except for those emitted by the dye in the sample.
