Click here to enter text. 5. What is the relationship between the working distance of an objective lens and its magnification power? Click here to enter text. 6. In what way(s) can you control the amount of light reaching the objective lens? Click here to enter text. 7. List the steps to correctly putting away a microscope. Click here to enter text. 8. What effect does increased magnification have on the field of view? Click here to enter text.

Microscopy MICROSCOPY PRE-LAB QUESTIONS 1. Of the four major types of microscopes, give an example of a scenario in which each would be the ideal choice for visualizing a sample. 2. What is the difference between the coarse adjustment knob and the fine adjustment knob? When is it appropriate to use each of them? 3. If you are using a compound microscope with an ocular lens with a magnification of 10x and an objective lens with a magnification of 40x, what is the total magnification? 4. In this lab, we discussed preparation of a wet mount slide. Research and describe another slide preparation and an example of when it would be used.

Activity 3A: Properties of the Microscope This simple activity will help you understand the attributes of the microscope. It is imperative that you actively participate and ask questions when you have problems. We will be using the microscope often in lab and it will be easier for you if you learn the basics first. 1. A specimen viewed with the microscope is magnified by the eyepiece lens (10x) and the objective lens in alignment with the light pathway. You will be asked to create sketches for specimens viewed at specific magnifications (see boxes on the following page). To calculate total magnification, multiply the eyepiece magnification

Hands-On Microscope What observations do you make observing the letter āeā at low power? At what objective magnification can you no longer see the entire letter when looking through the ocular lens? List the objective lens magnifications you used to observe the letter āeā below, along with your observations at each magnification. What did you use to create your second microscope slide? Write down your observations in viewing your second slide, and write down what magnifications you used to make those observations. Reflection (Discuss what you have learned by doing this experiment. How have your ideas changed? Do you have any new questions? What connections did you make between the lab and lecture?):
