Procedure 2.4 SUDAN IV TEST FOR LIPIDS Sudan IV is a red dye that dissolves in lipids. A positive result shows the formation of red droplets or, in some cases, a floating red layer appears. To perform the test, obtain four test tubes and number them with a wax pencil. Carry your test tubes in a test tube rack. Add the materials listed in Table 2.4. After you have added the food substances, add 2 ml of water to each test tube. Then, add five drops of Sudan IV to each tube. Mix the contents of each tube. It may be helpful to heat each tube in the heat block for several minutes (2-3 minutes). Let the test tubes sit for several minutes. Observe the final color and record it in Table 2.4. If there is a lipid present, the red color will appear at the top of the liquid, or you will see red particles or droplets.
