Put in the correct order the following steps of the Ras-MAPK signaling pathway downstream of a receptor tyrosine kinase. 1. Ligand binding results in dimerization of the enzyme-linked receptor. 2. Phosphorylated tyrosine residues on the receptor recruit Grb2 through its SH2 domain. 3. Grb2 uses its SH3 domain to recruit the GEF, Sos. 4. SOS activates Ras. 5. Ras activates MAP kinase-kinase-kinase (raf). 6. MAP kinase-kinase-kinase phosphorylates MAP kinase-kinase (mek). 7. MAP kinase-kinase phosphorylates MAP kinase (erk). 8. MAP kinase phosphorylates & activates downstream molecules such as transcription factors & cytoskeletal proteins. 9. Kinase domains on the receptor cross-phosphorylate the cytoplasmic tail of the partner.

Receptor tyrosine kinases (RTKs) can activate multiple signaling pathways. The PI3 kinase pathway is diagrammed below: You express green fluorescently labeled Akt (GFP-Akt) in cells. Under which of the following conditions would you expect to see fluorescence in the cytoplasm? Choose all that apply Bad mutant that cannot be phosphorylated; with ligand Wild-type cell; with ligand Gain of function PI3K mutant; no ligand Akt mutant missing the PH domain; with ligand Loss of function PI3K mutant; with ligand Receptor with tyrosines (Y) mutated to phenylalanine (F), with ligand

Place in order the following steps for the normal activity of a Ras protein. (1) Ras turns on the MAP kinase cascade. (2) Adaptor proteins connect phosphorylated tyrosine on a receptor to Ras. (3) GTP activates Ras by binding to it, displacing GDP. (4) The last MAP kinase in the cascade phosphorylates proteins in the nucleus that activate genes. (5) Receptor tyrosine kinase binds a signal molecule and is activated. a. 1, 2, 3, 4, 5 b. 2, 3, 5, 1, 4 c. 5, 2, 3, 1, 4 d. 2, 3, 1, 5, 4 e. 4, 1, 5, 3, 2
Place the following events involved in a receptor tyrosine kinase signaling pathway in the correct order/sequence: A. Transcription factors in the nucleus increase the expression of genes B. Receptors phosphorylate each other C. MAPK is activated D. Ligand binds to the receptors E. Receptors dimerize in the plasma membrane F. SOS-GRB2 complex binds to the receptor complex G. Ras has GTP bound H. Ras binds to the SOS-GRB2 complex
