The success of the anti-CD20 monoclonal antibody rituximab for rheumatoid arthritis prompted studies to test whether individuals with other diseases might benefit from rituximab treatment. What diseases would be good candidates for testing the therapeutic benefits of rituximab?
Added by Teresa P.
Step 1
Rituximab targets CD20 on B-cells, so diseases where B-cells play a significant role in pathogenesis are prime candidates. Show more…

A 27-year-old white woman sought medical attention because of persisting pain in her wrists and ankles and an unexplained skin irritation on her face. On physical examination, swelling of the joints of the hands and ankles was evident, along with erythema of the skin over the bridge of the nose and the upper cheeks. The patient had a slightly elevated temperature. Laboratory Data Complete blood count, urinalysis (UA), and rheumatoid arthritis (RA) screening test were ordered, with the following results: - Hemoglobin and hematocrit—normal - Total leukocyte count—70 × 10^9/L - Differential leukocyte count—normal - Gross and microscopic UA—normal - RA screening test—positive Follow-Up An ANA screening test was ordered. The results were positive. Group Discussion Questions 1. What is the most probable diagnosis in this case? 2. Does this patient fit into the general characteristics of patients with this disease? 3. What is the principle of the ANA test?

Humanized antibodies are currently being used as therapeutic agents for a number of infectious diseases, autoimmune diseases, and inflammatory diseases. These antibodies are often generated by grafting the critical amino acid residues of a mouse antibody, that exhibits the desired specificity, onto the backbone of a human IgG antibody. In one case, a mouse antibody with potent neutralizing activity against hepatitis B virus was the starting point, and sequences from this antibody were transferred onto the backbone of a human IgG. The ultimate goal of this antibody engineering effort was to produce a human IgG-based protein that could be administered to patients suffering from hepatitis B virus infection. The minimal sequences from the mouse virus-specific antibody that would be transferred would likely include the mouse antibody VH and VL framework sequences Fab sequences VH and VL CDR1, CDR2, and CDR3 sequences VH and VL CDR3 sequences

Rheumatoid arthritis is a disease that occurs when the body's immune system starts attacking its own joint tissue. The symptoms are pain, inflammation, and swelling in the joints. Orthopedists (doctors who study bones) and immunologists (doctors who study the immune system) both work with patients who have rheumatoid arthritis. What is the difference between the orthopedists and immunologists who treat the disease? Only the orthopedists examine the symptoms. O Only the immunologists examine the symptoms The orthopedists focus on the joints, and the immunologists focus on the cause The immunologists focus on the joints, and the orthopedists focus on the cause
