Under the Microscope A surgical technologist working in the Central Sterile Processing Department is asked to run the steam sterilizer (autoclave) with a biological monitor for the first load of the day without any instrument trays or items. 1. Which type of bacteria would be used to test the autoclave? 2. Which characteristic of the species used determines whether sterility was achieved when autoclaved? 3. What must be done with the bacterial sample after it is processed in the sterilizer? 4. Do the bacteria used for biological testing of sterilizers pose any threat to humans? 5. If the biological indicator showed positive growth following the process in question 3, what would have to be done?

Step 1
The bacteria used for testing autoclaves is typically *Geobacillus stearothermophilus*. This species is chosen because it is highly resistant to heat, making it an ideal candidate for testing the effectiveness of steam sterilization. Show more…

1. What are the reasons for using aseptic technique? 2. When setting up an experiment using bacteria, what materials must be sterilized or disinfected? How would you accomplish this for each of the materials? 3. Why does aseptic technique involve working close to a Bunsen burner flame? 4. What is the proper way to open a sterile serological pipet? What are two reasons that the pipet must not touch your hands, clothes or lab bench? 5. What is the purpose of streaking a plate using quadrants? What techniques can be used to result in decreasing bacterial growth in each quadrant? 6. Describe the concept of serial dilution of bacterial culture. When would you use this technique?

Which best describes the in-use test used to estimate disinfectant effectiveness? Microbe-contaminated stainless steel rings are immersed in a disinfectant for 10 minutes, then cultured to see if any of the bacteria survived. Disinfectant is placed on small paper discs, which are then placed on agar plates containing a lawn of bacteria to measure how effective the disinfectant is at killing the bacteria. Disinfectant is placed in a 2 mm well on an agar plate that has a lawn of bacteria covering the plate to measure how many of the bacteria are killed. Serially diluted disinfectants are inoculated with microbes and then cultured to check for bacterial growth. Cultures are taken from a disinfectant used in a hospital to see if any bacteria are growing in it.

A geneticist is interested in determining the locations of methylated cytosines within a fragment of DNA. She treats some copies of the fragment with sodium bisulfite and leaves some copies untreated. She then sequences the treated and untreated copies of the fragment and obtains the following results. Give the original sequence of the DNA frag ment and indicate the presence of methylated cytosines. Sequence without treatment: -AATTGCCCGATCGATTAAGCCA- Sequence with treatment: -AATTGTTTGATCGATTAAGCTA-
