Applications of Genetics: Imagine you are a genetic engineer and want to modify a deadly toxin (protein X) from AConor with the ultimate purpose of using it to cure brain cancer. You are given a small gene tube that codes for this protein X and you have modified it for treatment purposes. The protein X requires an increased concentration when used for recombinant DNA technology. Use this information to answer the following questions: 1. List each of the seven steps involved in rDNA technology, from obtaining large amounts of this gene to confirming the purity of the protein gene from an E. coli bacteria. Briefly describe one of them (2 pts). 2. The restriction enzyme mapping of the linear DNA reveals the following HindIII restriction sites: - HindIII site 1 - HindIII site 2 - Protein X gene - 270 - 645 - 700 This piece of DNA is cut with the indicated restriction enzyme and the resulting fragments are subjected to PCR amplification of the protein X gene. The amplified fragment is then separated by gel electrophoresis and stained for visualization. Find the length of the gene and draw a picture of the band that will appear on the provided gel for your sample, as well as for the positive and negative controls.
