What are the considerations for choice of a vector in gene therapy?
1. The cauliflower mosaic virus genome, when used as a vector, will not be able to express the aphid transmission protein if there is successful insertion of the gene of interest. 2. Viral vectors can be used to transform plant cells because plants have natural viral pathogens that transfer their genome to the plant. 3. Plant cells attain a protoplastic state only after removal of the cell wall. 4. Transient transfection is preferred if the gene of interest is desired to be expressed by generations of stable cell lines.

What is a feature peculiar to adeno-associated viral (AAV) vectors and not shared with retroviral vectors? Therapeutic gene integrates randomly into the patient's genome and could lead to cancer. Packaging the therapeutic DNA into virus-like particles requires "helper DNA" which expresses all the proteins required to make viral particles. Therapeutic gene is transcribed and translated in patient cells using the host transcription and translation machinery. Packaging cells do not make active viruses because helper DNA lacks packaging sequences and therapeutic DNA lacks essential viral genes. Gene therapy needs to be repeated periodically to provide a constant supply of the therapy protein.
