This identifies the charge on the resin. Cation exchange has negative resin... The wash buffer needs to have a pH and you need to carefully plan which proteins are stuck to the column and which is rinsed through. Then the elution buffer has a different pH to change the charge on the stuck protein. Which fraction you would keep and which is the contaminant. Same for the gel filtration (first or second) you need to state which fraction is the contaminant and which is the target.
