The survival and continued growth of microorganisms depend on an adequate supply of nutrients and a favorable growth environment. A solution containing these nutrients is a culture medium. A liquid medium lacks a solidifying agent and is called a broth medium. A broth medium with a solidifying agent called agar results in a solid or semisolid medium. Agar is an extract of seaweed. Agar liquefies at 100°C and solidifies at approximately 42°C. You will be preparing Trypticase Soy Broth (TSB) and Trypticase Soy Agar (TSA). Broth: 5 ml broth dispensed into small (#16) test tubes. Agar Slants: 10 ml of agar dispensed into large (#18) test tubes. After sterilization, the tubes are allowed to cool at an angle creating a slant. Agar Deeps: 15 ml of agar dispensed into large (#18) test tubes. Large tubes (#18) 18 mm x 150 mm. Small tubes (#16) 16 mm x 125 mm. CALCULATIONS. Each team is to prepare the following: Trypticase Soy Broth @ 5 ml/tube (#16 tubes). Trypticase Soy Agar FOR SLANTS @ 10 ml/tube (#18 tubes). Trypticase Soy Agar FOR DEEPS @ 15 ml/tube (#18 tubes). Each team will be assigned the number of tubes of each type of media to prepare: Broth: ___ tubes @ 5 ml = ___ ml broth. Agar: ___ tubes @ 10 ml = ___ ml agar (slants). Deeps: ___ tubes @ 15 ml = ___ ml agar (deeps). TOTAL ___ ml agar. Manufacturer's directions are for re-hydrating dry media: Trypticase Soy Broth: 30 gm dry media per 1,000 ml distilled water. Trypticase Soy Agar: 40 gm dry media per 1,000 ml distilled water.
