The ability to produce proteins through genetic engineering of microbial and mammalian cells and the need for high-purity therapeutic proteins has established a need for efficient large-scale protein purification schemes. The system of continuous affinity-recycle extraction (CARE) combines the advantages of well-accepted separation methods, such as affinity chromatography, liquid extraction, and membrane filtration, while avoiding the drawbacks inherent in batch and column operations. The technical feasibility of the CARE system was studied using ̢-galactosidase affinity purification as a test system. The figure below shows the process. What is the recycle flow rate in milliliters per hour in each stream? Assume that the concentrations of U are equivalent to the concentrations of the ̢-galactosidase in solution, and that steady state exists.

A recombinant protein is produced on pilot scale using E. coli. The bioreactor is operated on fedbatch mode under glucose-limiting conditions. The glucose concentration in the initial batch solution of 8 litres is 10 g/L. during the fed-batch phase 6 L of a concentrated glucose solution (410 g/L) are added under controlled conditions. The final biomass concentration is 50 g/L (dry cell weight basis) and the final protein product concentration is 10 mg/g biomass (dry cell weight). (i) Estimate the biomass yield coefficient on glucose, YX/S (g biomass/g glucose). (ii) Estimate the protein yield coefficient on glucose, YP/S (g protein/g glucose).
