Why are serial dilutions used with both the pour plate and spread plate methods of plating? Without dilution, the colonies would not be separated from one another and they would be difficult to count and identify. Without dilution, most of the bacteria would die before the colonies formed on the agar. Using serial dilutions concentrates bacteria so that they can form colonies. Using serial dilutions makes it possible to identify the nonliving cells in the original sample. It does not harm temperature-sensitive microbes.
