Step 1
- The first limitation of using the Hanging-drop method to observe microorganisms is the potential for rapid drying out of the specimen. This method involves placing a drop of culture or specimen in a depression slide, covering it with a cover slip, and then Show more…

Describe two benefits to producing wet mount slides as opposed to other methods of microscopy.

Describe the wet mount method for fungi. 1. Sterile techniques 2. Excavate an edge colony, trying to minimize the amount of agar. 3. Transfer to a slide, washing off the colony with 70% ethanol. 4. Tease out the mycelium with a wire loop. 5. Add a drop of lactophenol cotton blue and cover with a coverslip. 6. Blot excess dye. 7. Microscopy. 8. Why is 70% ethanol used in the wet mount?
Madhur L.
What is the advantage of observing a hanging drop, compared to a wet mount preparation? The resolution of the microscope lens is improved when viewing a hanging drop preparation. Motility and larger organisms are better viewed in a hanging drop due to the greater depth. A hanging drop preparation is easier to put together. A wet mount will last longer since the water will not evaporate as quickly.
