A batch of trichrome-stained slides for ova and parasite examination contains numerous minute crystals, which totally obscure the microscopic field. Which of the following measures is the most appropriate remedial action?a. change the Schaudinn fixative, remove coverslips and restainb. change the acid alcohol and restainc. remove coverslips and remount using fresh Permount or similar mediumd. change the iodine alcohol solution to obtain a strong tea-colored solution, restain

A small clinical laboratory collects specimens for parasitology examination but ships these specimens to a reference laboratory 250 miles away for the actual analysis. Which stool preservatives can be used for routine parasite study that consists of examination of concentrated sediment and trichrome-stained smears? Why? Which stool preservative yields the fastest turnaround time? Why? Discuss the pros and cons of performing both types of blood smears (thick and thin) on the same slide as opposed to making separate slides for each smear preparation. What is the source of refractive granules seen in a trichrome-stained smear of feces? How could the problem be resolved? During the trichrome staining procedure, the xylene becomes cloudy as the slide is introduced. Propose a source for the cloudiness and describe the action that should be taken to obtain acceptable results with this slide and those to follow.

You are viewing a prepared slide of striped muscle fibers from a cockroach leg. When you focus the microscope, the striations appear pale and distinct. To make the striations clearly visible, you would: a) Slowly close the diaphragm to reduce light. b) Remove the mirror to cut out light. c) Change the eyepiece to increase magnification. d) Replace the objective to decrease magnification.
Stain the specimen on a slide using the following procedure: 1. Fix in absolute alcohol. 2. Transfer sections to 90%, 70%, 50%, 25% alcohol, and finally water. 3. Place a drop of hematoxylin. 4. Transfer to acid alcohol to destain. 5. Rinse in ammonia. 6. Place in 70% alcohol. 7. Counterstain with eosin for 1-2 minutes. 8. Dehydrate in 70%, 90%, and 100% alcohol. 9. Clear in xylol. 10. Mount in glycerol. 11. Observe the specimen under the light microscope. 1. What is the function of xylol? 2. What is the meaning of destaining? Note: When staining the specimen, it should be in the same medium as the stain (if the stain is in water, the specimen should also be in water).
