A three-base sequence on a transfer RNA molecule that corresponds to a specific codon on mRNA and thus matches each amino acid with a specific codon. During translation, this area of a ribosome binds a tRNA molecule to which the growing polypeptide chain is attached. This protein binds to the A-site of a ribosome when a stop codon is reached, and then hydrolyzes the bond between the tRNA in the P-site and the polypeptide attached to it. This term describes the movement of a ribosome from one codon to the next in the 5' to 3' direction.
