Step 1
Step 1: Identify the initiating nucleophile for the first step of intron splicing: it is the 2'-hydroxyl group of the branch point A-residue that attacks the 5'-splice site. Show more…

What chemical moiety on the pre-mRNA initiates the removal of introns during alternative splicing reaction that ultimately forms the RNA lariat structure? A. the 2'-hydroxyl of the intronic adenine residue B. the amino group of the intronic adenine residue C. the oxygen of the phosphodiester group D. The 3'-hydroxyl of the intronic adenine residue

5'ss bp 3'ss pre-mRNA pppGAUAGCUAGCUAUGUAAGUCAUGAUACUAACUCUCAGACAUGCUAGCGGACUACGCUA... Above is shown the 5' end of a human pre-mRNA. The 5' splice site (5'ss), branch point (bp) and 3' splice site (3'ss) of the pre-mRNA's intron are indicated. 1a) As you know, pre-mRNA splicing requires U1, U2, U4, U5 and U6 snRNAs, and some of these base pair with the pre-mRNA to initiate splicing. Assuming that those snRNAs that base pair with the pre-mRNA each form 6 base pairs, box the nucleotides on the pre-mRNA sequence above that the snRNAs base pair with, clearly indicating the involved snRNAs. [Your boxed regions do not have to be exactly correct, but they have to make sense with the splicing reaction]. (4) 1b) Below is shown a representation of the intron as it looks after it has been spliced out of the pre-mRNA above. The four circles (i-iv) indicate positions of specific chemical groups. For each type of chemical group (phosphate or OH) listed below on the left, list in which of the four circles (i, ii, iii and/or iv) shown in the lariat intron they are found. For phosphate/OHs that are not found in any circle, write ‘None’. (4) Phosphate/OH Circle (i, ii, iii and/or iv, or None) 5'-3'-linked phosphate 5'-2'-linked phosphate 3'-2'-linked phosphate 5'-5'-linked phosphate 3' OH 5' OH Unlinked 3' phosphate Unlinked 5' phosphate 1c) As you know, the process of capping adds a GTP to the 5' end of the mRNA. On the sequence on the top of the page, box the phosphates of the pre-mRNA 5' end that remain part of the mRNA after capping. (2) 1d) You have identified three fungal toxins that all prevent the addition of GTP to the mRNA 5' end during capping. List three different proteins that could be targets of the three toxins. If you do not remember the specific name of the protein, in one sentence explain the activity of the protein. (3) 1: 2: 3:
