Experiment #1 – Selective and Differential Media (8 pts.)
A photograph your plates after incubation is provided below. Each plate has been labeled with the media type, incubation temperature, and bacteria. Refer to these plates when answering the following questions.
Figure 1. Lab strains inoculated on (A) TSA plates and (B) EMB plates incubated at 37°C for 24 hours. Inoculated strains are as follows; sector 1 - E.coli, sector 2 - Enterobacter aerogenes, sector 3 - Serratia marcescans, sector 4 - Bacillus cereus, sector 5 - Micrococcus luteus.
1) List the ingredients in EMB that enable the media to be differential and selective and explain why each works. (3 pts.)
2) For each strain on your EMB plates, characterize the appearance of the colonies that grew. Do you see different types of colonies? Describe the appearance of each of the test strains on your plates and explain what the presence of growth and color indicates about the type of bacteria present. (3 pts.)
