For each of the techniques, as succinctly as possible explain the What, When, and How. Example: When: When do you use this technique. What: What is the core of the technique. How: The rationale/tools that make it possible. Sticking with PCR: When: You want to amplify a certain stretch of DNA. What: Using cycles of the 3-step process of denaturation, annealing, and elongation that double the amount of DNA molecules from the previous cycle. How: You need a machine that can change temperature and a DNA polymerase, primers, and nucleotides to be able to perform each cycle till you are left with millions of DNA molecules.
