Name: Lab Section: Snyder test and bacteria of the mouth Fill in the blank: a simple and fun procedure that is used to evaluate a person's susceptibility to dental caries. The Snyder test is designed to favor the growth of oral bacteria (favor) and the Snyder test medium is formulated to discourage the growth of other bacteria, lowering the pH of the medium to pH 4.8. This is accomplished by adding glucose, which is fermented by bacteria in the mouth to form acid. Decalcification of enamel begins at pH below 5.5. Sucrose is broken down by bacteria in the mouth to form acid, which initiates dental caries by eroding the enamel. Only members of the Streptococcus genus and species survive these harsh conditions and also increase the pH even further. Streptococcus mutans is the most likely bacteria to cause dental caries. This bacterium is able to form a capsule made out of sucrose that allows it to adhere to the surface of the teeth. Masses of bacteria adhering to the teeth constitute dental plaque. You can easily tell the pH has decreased because the media has a pH indicator called bromothymol blue, which is green at or above pH 4.8 and turns yellow below pH 4.8. Development of color in this medium is evidence of fermentation and is highly suggestive of the presence of dental decay-causing bacteria. The media is maintained in a warm water bath to keep it liquid until needed. The agar is inoculated with bacteria and incubated up to 48 hours. The inoculated tube is incubated at 37°C, which is the optimal temperature for the growth of oral bacteria. If the person has high susceptibility to dental caries, the agar will change to yellow within 48 hours.
