Inoculation technique: stab & fishtail streak is important. It is not to touch the slant before stabbing the butt! All four tubes are incubated. Answer the following questions: 4. Why are we using an inoculation needle instead of a loop? 5. Why is it insufficient to streak the agar slant only? 6. Why is it important not to exceed 24-hour incubation when interpreting the results? 7. TSI is not a selective medium. Why is that irrelevant for our experimental design? If a loop is used to inoculate a tube of TSI agar, splitting of the agar can occur when the loop is stabbed into the media. In what way could this mechanical splitting of the media affect your evaluation of the tube after incubation?
