whats the difference between wet-mound and hanging drop preparation
Added by Lisa B.
Step 1
Wet Mount Preparation: This is a technique usually used to view live microorganisms. In this method, a drop of water is placed on a slide (sometimes with the addition of stains), the specimen is added to the water, and then it is covered with a cover slip. The Show more…

What is the advantage of observing a hanging drop, compared to a wet mount preparation? The resolution of the microscope lens is improved when viewing a hanging drop preparation. Motility and larger organisms are better viewed in a hanging drop due to the greater depth. A hanging drop preparation is easier to put together. A wet mount will last longer since the water will not evaporate as quickly.

What are the primary limitations imposed by using the Hanging-drop method (or Wet mount) to observe microorganisms? State at least two (2) accounts with a brief explanation. 2. What are the primary benefits of using the Hanging-drop method (or Wet mount) to observe microbes? State at least two (2) accounts with a brief explanation. 3. Based on the results of using the Hanging-drop method in Figure 2, what microorganisms can you find? State or identify all microbes you can see, and briefly describe each of them. 4. If you are going to actually observe and compare microscopically, the samples from a lake, canal/sewage, and tap water (from faucet), what possible microorganisms are common in all samples? State at least one (1) microbe and briefly explain why. 5. Why is it very important to have regular microbiological tests of water performed on natural bodies of water (river, spring, lakes, etc.), water treatment plants, industrial and residential tap water supply? Explain concisely your answer.

What can be observed in wet mounts or hanging-drop slides that cannot be observed in heat-fixed slides? What techniques are used to visualize and study the morphology of the viruses? Can you visualize and study the morphology of the viruses using simple techniques such as wet mounts, hanging-drop slides, heat-fixed slides?
