Why was a saline (0.9% NaCl) solution instead of distilled water used to remove cheek cells during a gargle rinse?
Why was Instagene, which contains Chelex (binds divalent ions), added to the tubes prior to boiling the buccal cells? (Hint: Eukaryotic cells contain many heavy metals such as Mg2+, Mn2+, Ca2+, etc.)
Cells were boiled to lyse cells. Why does this preparation not destroy the integrity of DNA for PCR use? Is DNA denatured? If so, what bond(s) is broken?
Why are the tubes centrifuged a final time before the pure DNA can be removed and stored into clean tubes?
