Nephron Loop Anatomy

The loop of Henle is divided into two segments—the descending limb and the ascending limb. Each has distinct structural and permeability characteristics that contribute to the kidney’s ability to concentrate urine. The descending limb is permeable to water but less active in solute transport, while the ascending limb is largely impermeable to water and actively transports solutes out into the medulla.

Transport Mechanisms in Renal Tubules

Different regions of the nephron employ different transport mechanisms. The descending limb primarily allows passive movement of water based on osmotic gradients, whereas the ascending limb engages in active transport to move ions such as sodium out of the tubular fluid, creating and maintaining the hyperosmotic medullary interstitium essential for water reabsorption in other nephron segments.

Cellular Energy Demand and Mitochondrial Concentration

Cells that perform active transport, particularly in the ascending limb of the loop of Henle, require significant amounts of ATP to power ion pumps. As a result, these cells have a high density of mitochondria. In contrast, cells lining the descending limb, which rely largely on passive water movement, have lower energy demands and thus fewer mitochondria.
